De Winne came to human spaceflight through military aviation and . His first spaceflight was the Belgian Odissea mission, launched on Soyuz TMA-1 and returned on Soyuz TM-34.
He later launched on Soyuz TMA-15 for 20 and 21, becoming the first European Space Agency astronaut to command the International Space Station.
After his flights, De Winne became a senior ESA human-spaceflight leader. His two Soyuz flights span both a short Belgian research mission and a long-duration ISS command assignment.
